Divergent genomic evolution in astrocytomas and oligodendrogliomas.

Divergent genomic evolution in astrocytomas and oligodendrogliomas.

In brief

Longitudinal whole-genome and whole-exome sequencing of tumors from 183 patients with IDH-mutant glioma shows astrocytomas and oligodendrogliomas follow divergent evolutionary paths. Recurrent oligodendrogliomas acquired PI3K and NOTCH1 mutations with higher mutational burden, while recurrent astrocytomas gained oncogene amplifications and CDKN2A/B homozygous deletions alongside chromosomal instability. In both, convergent acquired alterations tracked with worse outcomes, implying selection of treatment-refractory clones.

Why it matters

Defines genetically distinct evolutionary routes that may explain differential treatment response in IDH-mutant gliomas.
