Expert lecture explains how multiple Foxp3 enhancers cooperate to control Treg induction and stability
In brief
An expert lecture on ImmuneZoom argued that Treg induction efficiency is controlled cooperatively by multiple enhancers at the Foxp3 locus (CNS0, CNS2, CNS3 and others) rather than a single switch. Single enhancer deletions only mildly reduced Treg diversity and were tolerated, but combined deletions such as CNS0 plus CNS2, or CNS3 plus Aire, caused severe lethal autoimmunity; CNS0 and CNS2 double deletion also shortened mouse lifespan to 2-3 months. The speaker proposed a 'relative' tolerance model and said Foxp3 acts in dynamic complexes with factors such as AP1 and FOX, providing a theoretical basis for engineering Tregs for therapy.
