Deep dive

These three social media posts are all pre-congress promotional and traffic-driving content ahead of ESMO26, not data readouts from any trial. The congress details are explicit: ESMO26 will be held in Madrid, October 23-27. The first post, from Mednet, is a networking post naming three attendees - DocLHenry (Senior Medical Director, Oncology), Varsha Hotchandani (Head of Commercialization) and Travis Sullivan (Director, Business Development) - and inviting on-site meetings. The second, from touchONCOLOGY, is a retrospective collection of ESMO25 highlights, bringing together expert interviews covering ovarian, bladder, breast and oesophageal cancers as well as renal cell carcinoma. The third, from M3Community, is a pre-congress perspectives piece on RAS therapies, ctDNA, radioligand therapy, ADCs and AI.

As for which trial is actually being reported at this congress and its key numbers: none of the three posts discloses any specific trial name, tumor-stage detail, or efficacy and safety data; all relevant figures are undisclosed or to be confirmed. The touchONCOLOGY post explicitly points to last year's interviews rather than new data from this congress; the M3 post is a trends piece rather than trial results; and the Mednet post has nothing to do with trials. Any judgment on a specific ESMO26 readout, or on what step forward it represents relative to existing knowledge, cannot be made from this material set and must await the congress's official disclosures.

The background literature likewise contains no published pivotal trials directly corresponding to the RAS therapies, ctDNA, radioligand therapy or ADCs named in the posts, so class-level evidence within this material set remains to be confirmed. Only two pieces of general, tangentially relevant evidence are available: first, PD-1/PD-L1 blockade can develop acquired resistance through CD38 upregulation in the tumor microenvironment, and coinhibition of CD38 and PD-L1 improves the antitumor immune response [2]; second, the highly selective complex I inhibitor IACS-010759, in two phase I trials in relapsed/refractory acute myeloid leukemia (n=17) and advanced solid tumors (n=23), showed a narrow therapeutic index with dose-limiting toxicities including elevated blood lactate and neurotoxicity, no recommended phase 2 dose was established, only limited antitumor activity was observed at tolerated doses, and both trials were discontinued [3]. These two items can serve only as general background for understanding immune-resistance mechanisms and early development risk, and cannot be mapped onto any specific report at this congress.

The limitations are equally direct: this material consists entirely of pre-congress promotion, retrospective interviews and topic aggregation, with no primary data, abstract numbers or trial registration numbers; it can be used to track the congress agenda and topic momentum, but does not constitute a basis for judging any therapy or readout. All figures and conclusions must be verified after formal disclosure at the congress.
