Pharma’s $500B patent problem will come in waves — not as a cliff

Evaluate forecasting data from Norstella show more than $500 billion in worldwide prescription drug sales at risk from generic competition between 2026 and 2032, with the largest single-year exposure of $158 billion in 2031. Merck is most exposed near term as Keytruda nears expiry in 2028, a year with $99 billion in sales at risk, while AbbVie faces roughly $50 billion of exposure in 2032 when Rinvoq and Skyrizi lose protection. Patent-exposed companies have responded with dealmaking since the start of 2024, led by Merck's six deals totalling $30.9 billion and Lilly's $27.2 billion across nine.

Why it matters: Quantifies the multiyear patent-expiry risk facing major drugmakers and the dealmaking it is already driving.

Genmab and AbbVie’s Epkinly win, a rush of deals, Sarepta’s turnaround hopes, more

Genmab and AbbVie's bispecific antibody Epkinly, approved in May 2023 for diffuse large B cell lymphoma after at least two prior systemic lines, cut risk of disease progression or death by 51% in a Phase 3 trial when combined with R-CHOP in the frontline setting. In deals, Shionogi bought IntraBio for $2 billion, Novartis committed up to $7.8 billion to Abogen Biosciences, Eli Lilly added up to $870 million with Gate Bioscience, and Sanofi and Regeneron renewed their collaboration with a $1 billion upfront. Sarepta shared Elevidys data in older Duchenne patients at the World Muscle Society meeting, with Jefferies citing upcoming readouts and an FDA decision on Amondys 45 and Vyondys 53.

KRASG12C Inhibition Remodels the Non-Small Cell Lung Cancer Surfaceome to Uncover Targetable Vulnerabilities

KRAS G12C inhibitors remodel the cell surface proteome of KRAS G12C-mutant lung adenocarcinoma, and folate receptor alpha (FOLR1) emerges as a therapeutically actionable target that is significantly upregulated during treatment, part of an acute cellular state transition toward epithelial identity. Quantitative temporal proteomic profiling across cell lines, tumor models, and treatment durations identified this adaptation. Combining KRASi with mirvetuximab soravtansine-gynx, a clinically approved FOLR1-targeting antibody drug conjugate, enhanced tumor cell cytotoxicity and deepened response and tumor control in cellular and in vivo models.

AbbVie wins FDA approval for Parkinson’s drug from $8.7B Cerevel takeover

The FDA has approved tavapadon, branded Juvmo, for adults with Parkinson's disease, giving AbbVie a marketed asset from its $8.7 billion Cerevel Therapeutics acquisition. BMO calls Juvmo the first selective D1/D5 receptor agonist available in the U.S., usable with or without levodopa. AbbVie will list it at $4,900 per 30-day supply, or $58,800 per year, and expects U.S. availability next month, targeting a market opportunity the company puts at $5 billion.

Why it matters: Approval salvages a product from the Cerevel deal after emraclidine failed, and launches a first-in-class Parkinson's option.

Mid-sized biotechs pressured to accept MFN deals crafted for big pharma

A new round of most-favored nation drug pricing deals, expected to be announced as soon as Monday, reveals pressure from the Trump administration on mid-sized biopharmas. About 10 companies have accepted deals while others have resisted. The White House offers exemptions from mandatory Medicare MFN drug-pricing models and tariff relief in exchange for deep cuts to Medicaid reimbursement, with decisions hinging on payer mix and tariff exposure.

Efficacy and safety of upadacitinib in adults and adolescents for treatment of non-segmental vitiligo (Viti-Up): results of two phase 3 randomised controlled studies.

Upadacitinib 15 mg once daily produced clinically meaningful repigmentation in non-segmental vitiligo, a disease with no approved systemic therapies. In two phase 3 placebo-controlled trials, T-VASI 50 at week 48 was reached by 19% and 21% of patients on upadacitinib versus 6% on placebo in each study, and F-VASI 75 by 25% and 23% versus 6% and 7%; both coprimary endpoints met p<0·001. Safety was consistent with the known upadacitinib profile, with no new signals.

Why it matters: First phase 3 data supporting a systemic therapy for non-segmental vitiligo, an approved-therapy gap.
