Molecular mechanism of residue-specific nucleosome acetylation.

Molecular mechanism of residue-specific nucleosome acetylation.

In brief

Cryo-EM structures of the Drosophila MSL complex caught during catalysis show how histone H4 lysine 16 is acetylated in a residue-specific way: a stable product state directly visualizes the acetyl group transferred from coenzyme A to H4K16. Cooperative MSL subunits act as a molecular ruler conferring specificity, and recognition of H3K36 trimethylation speeds enzymatic activity, tying MSL function to active gene bodies and Drosophila dosage compensation.

Why it matters

It provides the structural basis for how a histone acetyltransferase achieves residue-specific H4K16 acetylation.
