Rapid Micronucleus-Sequencing (RaMN-Seq) Reveals Patterns of Bias in Induced and Spontaneous Chromosomal Instability

Rapid Micronucleus-Sequencing (RaMN-Seq) Reveals Patterns of Bias in Induced and Spontaneous Chromosomal Instability

In brief

RaMN-Seq is a roughly 2-hour, kit-style micronucleus purification method coupled to single micronucleus low-pass DNA sequencing for examining micronucleus contents. It recapitulates the known chromosome bias of nocodazole washout-induced mis-segregation in diploid cells, shows ruptured (cGAS-positive) and intact micronuclei have no substantial differences in chromosomal contents or sizes, and works in cancer cell lines and fresh-frozen tumour samples. It offers an inexpensive alternative to single cell genome sequencing for studying chromosomal instability.

Why it matters

A fast, low-cost method to profile micronucleus contents makes chromosomal instability analysis accessible in tumour samples.
