A non-canonical function of glutathione to suppress ferroptosis via FSP1.

Glutathione suppresses ferroptosis independently of GPX4, protecting GPX4 knockout cells through a pathway pinpointed by genome-wide CRISPR-Cas9 screening: ferroptosis suppressor protein 1 (FSP1). FSP1 uses GSH to generate reduced ubiquinone, which scavenges oxidized lipids, and inhibition of FSP1 abolishes this protection in vitro and in vivo. The pathway differs from NAD(P)H-driven ubiquinone reduction by not requiring FAD.

Why it matters: It uncovers a GPX4-independent route by which glutathione restrains ferroptosis, relevant to ferroptosis-related disease therapy.

The past and future impact of climate change on childhood malaria in Africa.

Human-caused climate change has had a small net effect on childhood malaria across sub-Saharan Africa: rising temperatures probably increased cases in East and southern Africa but averted a comparable number in West Africa, a net impact of 1 excess case per 1,000 children (95% CI -4 to 6). Projections to 2100 suggest climate change could avert 1 to 20 cases per 1,000 children continent-wide, but limiting warming from 3 °C to under 2 °C could prevent an average of 5 excess cases per 1,000 in high-elevation East Africa and southern Africa.

Why it matters: It settles a decades-old debate on whether climate change has already raised malaria burden in African children.

Structural basis for host membrane binding and remodeling by invading malaria parasites.

The endogenous structure of the Plasmodium falciparum moving junction, purified from invasion-stalled parasites, reveals a sailboat-shaped 1:1:1:1 complex of PfAMA1, PfRON2, PfRON4 and PfRON5. Two PfRON2 transmembrane helices anchor the complex in the red blood cell membrane, while PfRON2/4/5 form a basic platform that wedges seven amphipathic helices into the bilayer, indicating active host-membrane remodeling. Guided by the structure, a rationally designed small protein binder inhibits invasion.

Why it matters: First native structure of the malaria moving junction enables structure-based design of an invasion inhibitor.

Coordinated RNA- and protein-templated synthesis of double-stranded DNA by a dual reverse transcriptase immune system.

DRT3 antiviral immunity depends on cooperation between class 1 and class 2 reverse transcriptase activities that together produce self-complementary double-stranded DNA. DRT3a copies a 5'-ACACAC-3' RNA template to make poly-(dTdG) repeats, while DRT3b makes poly-(dCdA) repeats without any nucleic acid template, using active-site-adjacent residues as nucleotide gates, as shown by cryo-electron microscopy. DRT3 is toxic in cells lacking RecBCD, and the phage-encoded RecBCD inhibitor Gam triggers abortive infection, defining how complementary DNA synthesis drives defense.

Why it matters: It explains how two reverse transcriptases with distinct templating strategies cooperate to build antiviral double-stranded DNA.
